Mild X-linked Alport syndrome due to the COL4A5 G624D variant originating in the Middle Ages is predominant in Central/East Europe and causes kidney failure in midlife

Summary: In a study of children with X-linked Alport Syndrome in Poland, a single variant in COL4A5 was identified as the predominant cause, dating back to a founder effect in the 12-13th century. This variant, known as G624D, was associated with a milder clinical course compared to other pathogenic variants, but still conferred a significant risk for kidney failure and deafness. Males with the G624D variant are candidates for both existing and emerging therapies for Alport Syndrome.

Autosomal dominant tubulointerstitial kidney disease genotype and phenotype correlation in a Chinese cohort

Summary: Genes UMOD, HNF1B, MUC1, REN, and SEC61A1 are associated with autosomal dominant tubulointerstitial kidney disease (ADTKD). A study of 48 probands and their family members revealed potential associations between genotype and outcomes, with UMOD gene cysteine substitutions showing a better prognosis. Follow-up data suggested suspected ADTKD individuals had a better prognosis compared to ADTKD individuals.

Meta-analysis of GWAS for kidney function identified new loci and fine-mapped variants and genes, shedding light on the complex genetic architecture underlying kidney function. Fine-mapping also revealed specific associations with cystatin-based eGFR and blood urea nitrogen levels at multiple loci.
