Summary: Melatonin has been shown to inhibit RANKL-induced osteoclastogenesis by upregulating Rev-erb alpha expression and downregulating miR-882 expression. The miR-882/Rev-erb alpha axis may play a vital role in preventing osteoclastogenesis, suggesting that targeting Rev-erb alpha or inhibiting miR-882 could be potential mechanisms through which melatonin prevents osteoporosis.

Summary: Our study demonstrated that melatonin effectively blocked RANKL-induced osteoclastogenesis by inhibiting PRMT1 and ADMA expression, thus suppressing bone resorption. Additionally, melatonin inhibited the transcriptional activity of NF-κB by interfering with the binding of PRMT1 and NF-κB subunit p65 in RANKL-treated bone marrow-derived macrophages, mediating the anti-osteoclastogenic effect of melatonin.
