Rho GTPase regulatory proteins in podocytes

Jun Matsuda et al.

Summary: Rho GTPases play a crucial role in regulating the actin cytoskeleton and are implicated in the pathogenesis of proteinuric glomerular diseases. These GTPases are primarily regulated by three families of proteins – GEFs, GAPs, and GDIs – with their upstream regulatory mechanisms largely unknown in podocytes. Further studies are needed to bridge the knowledge gap in the field.

SRGAP1 Controls Small Rho GTPases To Regulate Podocyte Foot Process Maintenance

Manuel Rogg et al.

Summary: Previous research demonstrated the importance of small Rho GTPases in driving podocyte foot-process effacement, but a comprehensive understanding of their regulatory networks in podocytes is lacking. This study identified SRGAP1 as a key regulator of podocyte foot-process architecture, highlighting potential novel targets for intervention in glomerular diseases.

Generation of stress fibers through myosin-driven reorganization of the actin cortex

Jaakko Lehtimaki et al.

Summary: The study reveals that stress fibers can form without pre-existing actomyosin bundles, giving rise to structures known as cortical stress fibers, which are embedded in the cell cortex and assemble preferentially underneath the nucleus. The assembly process is orchestrated by non-muscle myosin II pulses, leading to the reinforcement of nascent focal adhesions and subsequent stabilization of the cortical stress fibers. These findings demonstrate a new mechanism for de novo generation of stress fibers from the actin cortex, highlighting the role of stochastic myosin pulses in the assembly of functional actomyosin bundles.

EPB41L5 controls podocyte extracellular matrix assembly by adhesome-dependent force transmission

Jasmin Maier et al.

Summary: EPB41L5 protein plays a critical role in regulating podocyte adhesome function, affecting extracellular matrix assembly and deposition of core GBM components. This study reveals how EPB41L5 influences cell-cell force transmission and ECM formation by controlling the maturation of integrin adhesion complexes.

Focal adhesion dynamics in cellular function and disease

Yasaswi Gayatri Mishra et al.

Summary: Cell-ECM adhesions, with integrins at their core, serve as a crucial link between the cell cytoskeleton and the extracellular matrix, playing a major role in cell signalling for mechanotransduction, migration, proliferation, and other cellular processes. The diverse integrin adhesion complexes, such as focal adhesions and invadosomes, are not only essential for normal cell survival and development, but also play significant roles in various pathological processes.

Tripartite Separation of Glomerular Cell Types and Proteomes from Reporter-Free Mice

Favian A. Hatje et al.

Summary: timMEP method successfully isolated mesangial cells, endothelial cells, and podocytes from mice, enabling protein-biochemical analyses without the need for reporters. The method revealed cell type-specific proteome differences between mouse strains and variations based on sex, age, and transgene, as well as highlighted a protein-based mechanism of podocyte injury in a specific animal model, showing the potential for comprehensive characterization of glomerular cell biology.
