Optimizing Coaxial Sonic Spray Geometry for Generating Water Microdroplets

Maria T. Dulay et al.

Summary: Sonic spray is a technique that produces a stream of neutral and charged microdroplets without the need for voltage, heating, laser irradiation, or corona discharge. By adjusting parameters such as water quality, water flow rate, nebulizing gas pressure, and sonic sprayer geometry, the size distribution and density of the microdroplets can be characterized. The smallest microdroplet diameters are obtained when the gas flow pressure is highest and there is no separation distance between the two capillary outlets.

Can electric fields drive chemistry for an aqueous microdroplet?

Hongxia Hao et al.

Summary: Reaction rates of common organic reactions can significantly increase by several orders of magnitude in aqueous microdroplets compared to bulk solution. However, the reasons for this rate acceleration are still poorly understood. By studying the electric field distributions at the air-water interface, it has been found that the electric field alignments along free O-H bonds at the surface are on average around 16 MV/cm larger than those in the interior of the water droplet. This difference in electric field distribution may contribute to the enhanced surface reactivity observed in microdroplets.

In-Source Microdroplet Derivatization Using Coaxial Contained-Electrospray Mass Spectrometry for Enhanced Sensitivity in Saccharide Analysis

Derik R. Heiss et al.

Summary: The online droplet-based in-source chemical derivatization method using a coaxial-flow contained-electrospray ionization source significantly enhances sensitivity for the mass spectrometric analysis of saccharides. By optimizing ion source parameters with a Design of Experiments approach, signal enhancements of greater than two orders of magnitude were observed, achieving picomolar limits of detection for many mono- and disaccharides. This new approach can expand the utility of ESI-MS analysis for compounds with historically limited sensitivity and detectability, while minimizing resource-intensive derivatization procedures.

Incorporating Electro-Epoxidation into Electrospray Ionization Mass Spectrometry for Simultaneous Analysis of Negatively and Positively Charged Unsaturated Glycerophospholipids

Shuli Tang et al.

Summary: A new method utilizing AC-induced electroepoxidation reaction and nanoelectrospray ionization was developed to locate carbon-carbon double-bonds in lipids with positive and negative charges simultaneously. The AC voltage played multiple roles in initiating electro-epoxidation and protonation/deprotonation of lipids. This method was effective in lipid standards mixtures and a biological polar lipid extract, showing potential for wide applications in lipid-related research.

Microdroplet Ultrafast Reactions Speed Antibody Characterization

Pengyi Zhao et al.

Summary: Microdroplet reactions have shown to be an effective method for accelerating large protein reactions, particularly for the digestion, reduction, and deglycosylation of intact antibodies. By introducing appropriate enzymes and reagents into the spray solution, these multiple-step reactions can be completed rapidly, allowing for fast structural characterization of antibodies. This study demonstrates the potential of microdroplets as powerful microreactors for expediting large-molecule reactions and facilitating their structural analysis.
