Broken-Symmetry Density Functional Theory Analysis of the Ω Intermediate in Radical S-Adenosyl-L-methionine Enzymes: Evidence for a Near-Attack Conformer over an Organometallic Species

Summary: A study using density functional theory evaluated several structural models of Omega and found that the proposed organometallic structure is inconsistent with experimental results. Instead, a near-attack conformer of SAM bound to the [4Fe-4S] cluster was found to be more consistent with experimental results, suggesting a new understanding of the structure of the ubiquitous Omega intermediate and the mechanism of SAM cleavage by members of the radical SAM superfamily.

Summary: Research demonstrates that S-adenosyl-l-ethionine (SAE) can serve as an alternative to SAM in supporting the enzymatic turnover of HydG and forming organometallic intermediates. Photolysis of SAE-bound HydG results in the formation of an ethyl radical that can be stored at low temperatures for extended periods. The ethyl radical, when heated above 77 K, adds to the [4Fe-4S](2+) cluster, generating an ethyl-[4Fe-4S](3+) organometallic species.

Summary: Research has found that typical RS enzymes exhibit two types of S-C bond cleavage under light-induced conditions, depending on ribose conformation rather than the position of the sulfonium center relative to the [4Fe-4S] cluster. Additionally, the cleavage of the S-C bond results in selective cleavage, leading to the generation of radicals.

Trapping a cross-linked lysine-tryptophan radical in the catalytic cycle of the radical SAM enzyme SuiB

Summary: Using electron paramagnetic resonance (EPR) spectroscopy, this study dissected the mechanism of SuiB and revealed the essential role of the auxiliary [4Fe-4S] cluster in catalysis, confirming the proposed radical electrophilic aromatic substitution (rEAS) mechanism for the formation of the Lys-Trp cross-link. The findings provide direct evidence for the involvement of the SPASM domain auxiliary cluster and support rEAS as the mechanistic paradigm for rSAM enzyme-catalyzed carbon-carbon bond-forming reactions.
