α-Synuclein arginylation in the human brain

Jun Zhao et al.

Summary: In this study, the arginylation of alpha-synuclein (alpha-syn) was found to occur at two sites, E46 and E83, in human brain tissue. The level of arginylation was inversely correlated with overall alpha-syn levels and patient age, suggesting a potential neuroprotective role of alpha-syn arginylation in preventing abnormal accumulation during neurodegeneration and aging in the human brain.

Chemoenzymatic Semi-synthesis Enables Efficient Production of Isotopically Labeled α-Synuclein with Site-Specific Tyrosine Phosphorylation

Buyan Pan et al.

Summary: This study explores the impact of post-translational modifications on alpha-synuclein and introduces a streamlined method to produce phosphorylated alpha S in large quantities. By co-expressing a kinase with a protein fragment in Escherichia coli and using methyl thioglycolate, the researchers achieved high yields of site-specifically phosphorylated protein through chemoenzymatic methods. This strategy is particularly suitable for producing N-15-labeled, phosphorylated protein for NMR studies.

New strategies for fluorescently labeling proteins in the study of amyloids

Marie Shimogawa et al.

Summary: Amyloid proteins are extensively studied due to their unique biophysical properties and association with diseases like Alzheimer's and Parkinson's. Fluorescence-based methods using labeled proteins can offer insights into the structural dynamics and biological roles of amyloid proteins. Different labeling methods and fluorescent probe strategies are applied for in vitro biophysical experiments and in vivo imaging to study amyloid proteins and their pathology.

Consequences of post-translational modifications on amyloid proteins as revealed by protein semisynthesis

Stuart P. Moon et al.

Summary: Alterations in the global levels of certain PTMs are commonly observed in neurodegenerative diseases. However, the specific influence of these PTMs on individual proteins varies, requiring protein-specific investigations. Understanding how PTMs affect amyloid-forming proteins' behavior is crucial, and protein semisynthesis techniques have been widely used to study this relationship.
