A positive pressure workstation for semi-automated peptide purification of complex proteomic samples

Louise Schelletter et al.

Summary: The study evaluated a positive pressure workstation for increasing automation in sample preparation in proteomics. The use of a new semi-automated sample preparation device, Resolvex A200, with nanospray liquid chromatography/electrospray ionization (nLC/ESI) Orbitrap tandem mass spectrometry (MS/MS) produced highly reproducible results within a specified range of peptide starting material. The new workflow showed similar technical variability to the manual workflow, with successful application to a biological question and potential for further large-scale proteomic investigations.

Adding polyvinylpyrrolidone to low level protein samples significantly improves peptide recovery in FASP digests: An inexpensive and simple modification to the FASP protocol

Tammy-Lynn Tremblay et al.

Summary: Adding polyvinylpyrrolidone-40 (PVP-40) to protein samples before FASP digest significantly improves peptide recovery and identifications, especially with lower level sample inputs. PVP-FASP produces clean samples that require no additional clean-up steps before nanoLC-MS analysis.

Single-Cell Proteomics

Luke F. Vistain et al.

Summary: The inability to make broad, minimally biased measurements of a cell's proteome is a major bottleneck in understanding gene expression and cellular phenotype. Current methods for single-cell proteomic measurements typically focus on either absolute quantification of a small number of proteins or highly multiplexed protein measurements, with advancements in microfluidics and output encoding enabling major improvements. Emerging technologies like single-cell mass spectrometry may provide unbiased measurement of cellular proteomes.
