Beyond structural models for the mode of action: How natural antimicrobial peptides affect lipid transport

Josefine Eilso Nielsen et al.

Summary: By combining state-of-the-art small angle X-ray/neutron scattering techniques, this study found that various natural antimicrobial peptides accelerate essential lipid transport processes without altering the overall structure of lipid membranes, potentially triggering lethal signaling processes and enhancing ion transport.

Current approaches in lipid-based nanocarriers for oral drug delivery

Maria Plaza-Oliver et al.

Summary: Lipid-based nanocarriers have advantages like low toxicity and strong biocompatibility, making them ideal carriers for drugs with poor oral bioavailability. However, efficient oral drug delivery through these carriers is challenging due to multiple physicochemical barriers along the gastrointestinal tract. Rational design of nanocarriers can turn these limitations into opportunities, ultimately determining their future clinical success.

Stiffening Effect of the [Bmim][Cl] Ionic Liquid on the Bending Dynamics of DMPC Lipid Vesicles

Pallavi Kumari et al.

Summary: This study focuses on the effect of the model imidazolium-IL [bmim] [Cl] on the bending modulus of DMPC lipid vesicles, showing that the IL diffuses into the lipid region, accounting for five IL cations for every 11 lipids, and causes an increase of the lipid bilayer bending modulus, up to 60% compared to the neat lipid bilayer at 40 degrees C.

The distinct effects of two imidazolium-based ionic liquids, [C4mim][OAc] and [C6mim][OAc], on the phase behaviours of DPPC

Xiao-Lei Hao et al.

Summary: Ionic liquids are potential green solvents with broad application prospects, but their toxicity is largely related to their hydrophobic properties. This study explored the effects of two imidazolium-based ILs on the phase behaviors of a representative phospholipid. It found that [C(4)mim][OAc] had limited effects, while [C(6)mim][OAc] could significantly affect the lipid's main phase transition and form an interdigitated gel phase.

Computational analysis of the effect of [Tea][Ms] and [Tea][H2PO4] ionic liquids on the structure and stability of Aβ(17-42) amyloid fibrils

D. Gobbo et al.

Summary: Experimental studies have shown that the addition of organic salts can affect the growth/dissolution of amyloid fibers, with a stronger effect observed for [Tea][Ms]. Computational analysis revealed that both [Tea][Ms] and [Tea][H2PO4] decrease the stability of fibrils compared to isolated peptides in solution. The secondary structure of fibrils remains largely unaffected, but single peptides in solution show changes in beta-strand character and alpha-propensity, particularly with [Tea][Ms].

Time-resolved SANS reveals pore-forming peptides cause rapid lipid reorganization

Michael H. L. Nguyen et al.

Summary: This study investigates the effects of antimicrobial pore-forming peptides alameithicin and melittin on lipid exchange and flip-flop in model lipid vesicles. It was found that alameithicin increased both DMPC flip-flop and exchange by approximate to 2-fold, while melittin affected DMPC flip-flop by a factor of 1 to 4 depending on the concentration. Despite differences in their proposed oligomeric pore structures, both peptides showed remarkable similarity in their mechanism of action on lipid motion at high protein concentrations.
