Construction of Thioamide Peptide via Sulfur-Involved Amino Acids/Amino Aldehydes Coupling

Yanyan Liao et al.

Summary: A sulfur-involved ligation for thioamide quasi-peptides was developed via coupling of amino acids and amino aldehydes, with chelation of copper with imines for chiral activation and fixation being key to the transformation. Linear polysulfur was used to decrease the alkalinity of single sulfur anions, preventing racemization caused by interaction with sodium sulfide, resulting in successful synthesis of dipeptides, tripeptides, tetrapeptides, and drug-amino acid linkages.

Desolvation of Peptide Bond by O to S Substitution Impacts Protein Stability

Bhavesh Khatri et al.

Summary: Amino acid side chains play a key role in adjusting the polarity of the microenvironment in proteins, with substituting an oxygen atom with a sulfur atom in the backbone amide bond leading to increased lipophilicity. This single atom substitution significantly stabilizes proteins by reducing microenvironment polarity and enhancing protein stability through favorable van der Waals and hydrogen bonding interactions.

Allenone-Mediated Racemization/Epimerization-Free Peptide Bond Formation and Its Application in Peptide Synthesis

Zhengning Wang et al.

Summary: Allenone has been identified as a highly effective peptide coupling reagent for the first time, showcasing its robustness in forming peptide bonds. It is not only effective for synthesizing simple amides and dipeptides, but also suitable for peptide fragment condensation and solid-phase peptide synthesis (SPPS). This method represents a disruptive innovation in peptide synthesis by combining the advantages of conventional active esters and coupling reagents while overcoming their disadvantages.

Recent Advances in Ynamide Coupling Reagent

Tao Liu et al.

Summary: Coupling reagents can promote the direct condensation of carboxylic acids with amines or alcohols to form amide or ester bonds, widely used in drug, material, and cosmetic production. However, issues such as impurities and racemization caused by over activation of traditional coupling reagents, as well as large chemical waste from solid phase peptide synthesis, call for new reagents and mechanisms to address them.
