Summary: Dolastatin 10 is a highly potent broad-spectrum antitubulin anticancer pentapeptide isolated from Dolabellaauricularia. It can be modified at the C- and N-terminals to enhance its cytotoxic activity. However, clinical trials showed limited efficacy due to the lack of separation between toxicity and efficacy. Transitioning to antibody drug conjugates (ADCs) reduced systemic toxicity and several ADC drugs have been approved for cancer treatment.

Structural Diversity of Perylenequinones Is Driven by Their Redox Behavior

Summary: This study isolated and characterized the long-postulated biosynthetic precursor of class B fungal perylenequinones from a Shiraia-like sp. Furthermore, it investigated the structural relationship and redox behaviors of hypocrellins and hypomycins. The study found that hypocrellins can undergo reversible reduction and reoxidation under aerobic conditions, and can undergo ring closing metathesis under anaerobic conditions.

Additionally, PHY34 interacts with ATP6V0A2 subunit and induces cell death.

Targeting helicase eIF4A with flavaglines has been proposed as a pan-viral strategy for future RNA virus pandemics.
