Forkhead Transcription Factors in Health and Disease

Laetitia Herman et al.

Summary: FOX proteins belong to a conserved family of transcription factors that have evolved through gene/genome duplication, showing both functional redundancy and specific roles in development and adulthood. Genetic alterations or misregulation of FOX genes can lead to human genetic diseases, cancer, and/or aging. This review provides an updated overview of the characteristics of FOX family members in terms of expression, protein domain composition, evolution, and function.

Toward a mechanistic understanding of DNA binding by forkhead transcription factors and its perturbation by pathogenic mutations

Shuyan Dai et al.

Summary: FOX proteins are an evolutionarily conserved family of transcription factors that play regulatory roles in eukaryotes, with dysfunction implicated in various human diseases. Despite sharing a highly conserved DNA-binding domain, differences exist in the structure and DNA recognition mechanisms among FOX family members, calling for further systematic reviews.
