Novel Carboline Fungal Histone Deacetylase (HDAC) Inhibitors for Combinational Treatment of Azole-Resistant Candidiasis

Zhuang Li et al.

Summary: A series of novel carboline fungal HDAC inhibitors were designed and synthesized to combat antifungal drug resistance, with compound D12 showing potent synergistic effects with fluconazole against resistant Candida albicans infection. D12 reduced the virulence of C. albicans by blocking morphological mutual transformation and inhibiting biofilm formation, and reversed drug resistance by downregulating the expression of the azole target gene ERG11 and efflux gene CDR1.

Lanosterol 14α-demethylase (CYP51)/histone deacetylase (HDAC) dual inhibitors for treatment of Candida tropicalis and Cryptococcus neoformans infections

Tianbao Zhu et al.

Summary: In this study, novel CYP51/HDAC dual inhibitors were designed to show potent antifungal activity against Cryptococcus neoformans and Candida tropicalis infections, by inhibiting various virulence factors and down-regulating resistance-associated genes. This highlights the potential of CYP51/HDAC dual inhibitors as a promising strategy for the discovery of novel broad-spectrum antifungal agents.

Treatment strategies for cryptococcal infection: challenges, advances and future outlook

Kali R. Iyer et al.

Summary: Cryptococcus species, particularly Cryptococcus neoformans and Cryptococcus gattii, have a significant impact on global human health, with almost a quarter of a million cases and 181,000 deaths from cryptococcal meningitis annually. Despite limited treatment options and high mortality rates, only three major classes of drugs are approved for clinical use. Challenges in developing new therapies include host toxicity, pathogen resistance, and the limited efficacy of existing antifungal agents. Research efforts are focused on understanding resistance mechanisms, exploring novel therapeutic strategies, and accelerating drug discovery through genomics technologies and high-throughput screening.

Re-emerging Aspartic Protease Targets: Examining Cryptococcus neoformans Major Aspartyl Peptidase 1 as a Target for Antifungal Drug Discovery

Robin Krystufek et al.

Summary: Cryptococcosis is an invasive infection that contributes to 15% of AIDS-related fatalities, posing a significant challenge in treatment. The study identified a protease named May1 as a potential target for developing drugs that target both fungal and retroviral proteases, leading to the discovery of a potent inhibitor through combinatorial screening of compounds. This dual-specificity inhibitor shows promise as a lead compound for further development of inhibitors targeting May1 and HIV protease.

Discovery of sertraline and its derivatives able to combat drug-resistant gastric cancer cell via inducing apoptosis

Chao Mu et al.

Summary: The study found that the antidepressant drug sertraline could sensitize drug-resistant gastric cancer cells, and through the synthesis and evaluation of 30 derivatives, the activity of the best compound was significantly improved. Additionally, it was discovered that apoptosis induction and cell cycle arrest were the reasons for the death of drug-resistant cells, involving the PI3K/Akt/mTOR pathway.
