High-throughput screening assays for SARS-CoV-2 drug development: Current status and future directions

Tuan Xu et al.

Summary: In response to the COVID-19 pandemic, a panel of assays has been developed to screen drugs for anti-SARS-CoV-2 activity. The study found that multitarget assays were more accurate and efficient, while target-specific assays were better for investigating compound mechanisms of action. Additionally, strict filtering with counter screens may be more harmful than beneficial in identifying true positives, highlighting the importance of developing novel HTS assays against multiple targets in the SARS-CoV-2 life cycle.

Biological activity-based modeling identifies antiviral leads against SARS-CoV-2

Ruili Huang et al.

Summary: The biological activity-based modeling approach uses compound activity profiles to predict activity across different assays or targets, aiding in the discovery of potential antiviral drug candidates. This method successfully identified compounds with potential activity against Zika, Ebola, and SARS-CoV-2 viruses, demonstrating its effectiveness in predicting antiviral compounds. The confirmed anti-SARS-CoV-2 compounds show promise as potential therapies, with a focus on viral entry inhibition and autophagy modulation.

SARS-CoV-2 M-pro inhibitors with antiviral activity in a transgenic mouse model

Jingxin Qiao et al.

Summary: The study designed and synthesized 32 new M-pro inhibitors containing bicycloproline, which showed inhibitory effects on SARS-CoV-2. Compounds MI-09 and MI-30 exhibited excellent antiviral activity in cell-based assays and significantly reduced lung viral loads and lung lesions in a transgenic mouse model of SARS-CoV-2 infection. Both also displayed good pharmacokinetic properties and safety in rats.
