Single-molecule FRET imaging of GPCR dimers in living cells

Summary: This study presents a generally applicable method for using single-molecule fluorescence resonance energy transfer (smFRET) to detect and track transmembrane proteins diffusing within the plasma membrane of mammalian cells. The in-cell smFRET approach reveals agonist-induced structural dynamics within individual metabotropic glutamate receptor dimers. Evidence for receptor monomers, density-dependent dimers, and constitutive dimers in representative class A, B, and C receptors were observed using these methods.

NPY-Y1 receptor signaling controls spatial learning and perineuronal net expression

Summary: This study demonstrates that conditional depletion of the gene encoding the Y1R for NPY in adult forebrain excitatory neurons results in a significant slowdown in spatial learning, with an intensification of PNN expression and an increase in c-Fos expressing cells in the CA1. Importantly, enzymatic digestion of PNNs in CA1 normalizes c-Fos activity and rescues the learning abilities of Npy1r(rfb) mice, revealing a functional link between NPY-Y1R transmission and PNNs in the control of dorsal hippocampal excitability and cognitive functions.

Crystal Structure-Guided Design of Bisubstrate Inhibitors and Photoluminescent Probes for Protein Kinases of the PIM Family

Summary: The X-ray crystallographic study of PIM-1 protein kinase complexes with three inhibitors revealed simplified chemical structures with reduced polar groups and chiral centers, which showed low-nanomolar potency and high selectivity towards PIM kinases. The developed inhibitors were functionalized with biotin or fluorescent dye Cy5 for detection in biochemical solutions and complex biological samples. Utilizing a PIM-2 selective detection antibody, a sandwich assay was able to detect as low as 44 pg of active recombinant PIM-2.
