Large-Scale Multi-omic Analysis of COVID-19 Severity

Katherine A. Overmyer et al.

Summary: The study conducted RNA-seq and high-resolution mass spectrometry on blood samples from COVID-19-positive and COVID-19-negative patients, identifying molecular features associated with disease severity and outcomes, offering pathophysiological insights and therapeutic suggestions. The observed dysregulation of platelet function, blood coagulation, acute phase response, and endotheliopathy further illuminated the unique COVID-19 phenotype. The findings are presented through a web-based tool enabling interactive exploration and machine learning prediction of COVID-19 severity.

Von Willebrand factor propeptide in severe coronavirus disease 2019 (COVID-19): evidence of acute and sustained endothelial cell activation

Soracha E. Ward et al.

Summary: EC activation plays a key role in the pathogenesis of severe COVID-19, as evidenced by increased plasma VWF:Ag and FVIII levels in patients. Elevated levels of VWF-FVIII complex and VWFpp were sustained for up to 3 weeks, with a reduced VWFpp/VWF:Ag ratio indicating decreased VWF clearance. Importantly, plasma VWFpp levels correlated with clinical severity indices in severe COVID-19, suggesting a potential role as a biomarker in this setting.

The ADAMTS13-von Willebrand factor axis in COVID-19 patients

Ilaria Mancini et al.

Summary: Severe COVID-19 patients exhibit an increased risk of thromboembolic events, with evidence of microthrombosis in the lungs of deceased patients. The study found a significant alteration of the VWF-ADAMTS13 axis in COVID-19 patients, with an elevated VWF:Ag to ADAMTS13 activity ratio strongly associated with disease severity, enhancing the hypercoagulable state and risk of microthrombosis in these patients.

The interplay between inflammatory pathways and COVID-19: A critical review on pathogenesis and therapeutic options

Shalki Choudhary et al.

Summary: COVID-19, caused by SARS-CoV-2, leads to acute respiratory distress syndrome (ARDS) through inflammatory signaling pathways and cytokine storm. Excessive secretion of pro-inflammatory cytokines and dysregulation of the immune system can cause immune damage, while dysfunction of the renin-angiotensin system (RAS) is associated with mortality. There is currently no specific treatment available, but various treatment options are being researched by scientists.

The miRNA: a small but powerful RNA for COVID-19

Song Zhang et al.

Summary: This article summarizes the role of miRNA in COVID-19 infection and suggests that COVID-19 cleverly exploits the interaction between miRNA and other biomolecules to evade host immune response. The study proposes that the miRNA-based interplay between the host and SARS-CoV-2 may be the primary cause of SARS-CoV-2 accessing and attacking host cells.

MicroLet-7b Regulates Neutrophil Function and Dampens Neutrophilic Inflammation by Suppressing the Canonical TLR4/NF-κB Pathway

Binzhen Chen et al.

Summary: In this study, the regulatory pathway of miR-let-7b/TLR4/NF-kappa B in neutrophils was investigated. Overexpression of miR-let-7b down-regulated TLR4 expression and decreased proinflammatory factors while increasing anti-inflammatory factor in vitro. Treatment with miR-let-7b agomir in septic mice improved survival, reduced organ injuries, and inhibited neutrophil recruitment.

Acute and post-acute phase of COVID-19: Analyzing expression patterns of miRNA-29a-3p, 146a-3p, 155-5p, and let-7b-3p in PBMC

Tahereh Donyavi et al.

Summary: This study aimed to identify specific miRNA profiles that can be used as biomarkers to differentiate acute COVID-19 cases, healthy individuals, and those in the post-acute phase. The results showed that miR-29a-3p, miR-146a-3p, and let-7b-3p were significantly elevated in COVID-19 patients compared to healthy individuals, with potential for high sensitivity and specificity in COVID-19 diagnosis and monitoring. MiR-29a-3p and miR-146a-3p also showed promise as biomarkers for distinguishing acute from post-acute phases of COVID-19.
