Differential Modulation of the Central and Peripheral Monoaminergic Neurochemicals by Deprenyl in Zebrafish Larvae

Summary: The study found significant changes in the levels of ten monoaminergic neurotransmitters in the head, trunk, and whole body of zebrafish larvae after treatment with 5 M deprenyl. While most monoaminergic neurochemicals were higher in the head than in the trunk in control larvae, deprenyl treatment resulted in changes with serotonin and norepinephrine increasing in both the head and trunk, and dopamine, L-DOPA, and homovanillic acid levels being modulated only in the head. Notably, the highly significant increase in dopamine levels observed in the head after deprenyl treatment was not detected in the whole-body analysis, indicating that the analysis of neurotransmitters in the whole body of zebrafish larvae should not be used as a general surrogate of the brain.

Pharmacological Modulation of Serotonin Levels in Zebrafish Larvae: Lessons for Identifying Environmental Neurotoxicants Targeting the Serotonergic System

Summary: This study investigated the acute effects of pharmacological modulation of the serotonergic system on cognitive, basic, and defense locomotor behaviors in zebrafish larvae. The results showed that deprenyl and fluoxetine led to impaired locomotion and cognitive function, while PCPA induced hyperlocomotion and increased escape responses. Co-exposure to agonistic and antagonist serotonin signaling drugs partially restored impaired behaviors and gene expression in zebrafish larvae.

Androgenic activation, impairment of the monoaminergic system and altered behavior in zebrafish larvae exposed to environmental concentrations of fenitrothion

Summary: The study demonstrates that exposure to environmental concentrations of Fenitrothion can induce neurotoxic effects in zebrafish larvae, impacting their basal locomotor activity, visual-motor response, and acoustic/vibrational escape response. Significant non-monotonic concentration-response relationships were observed for Fenitrothion and the expression of certain genes. Furthermore, potential protein targets of Fenitrothion were identified, with interactions with acetylcholinesterase, monoamine-oxidases, and androgen receptor being experimentally validated, suggesting an environmental risk for fish communities.
