Gene body methylation facilitates the transcription of CTSG via antisense lncRNA AL136018.1 in dermatomyositic myoideum

Ying Liang et al.

Summary: The study found that antisense long noncoding RNA AL136018.1, adjacent to CTSG in dermatomyositis patients, is highly expressed in skeletal muscle tissues and positively correlates with the transcription and DNA methylation levels of CTSG. The longer transcript of AL136018.1, AL136018.1-201, binds to specific exons and introns of CTSG and recruits DNMT3a to facilitate DNA methylation and transcription of CTSG in gene body. This reveals a novel epigenetic mechanism regulating CTSG transcription in dermatomyositis.

LncRNA TINCR favors tumorigenesis via STAT3-TINCR-EGFR-feedback loop by recruiting DNMT1 and acting as a competing endogenous RNA in human breast cancer

Qin Wang et al.

Summary: The study revealed that TINCR is upregulated in human breast cancer tissues and its knockdown suppresses tumorigenesis in vitro and in vivo. Mechanistically, TINCR recruits DNMT1 to the miR-503-5p locus promoter, increasing methylation and suppressing transcriptional expression of miR-503-5p. Additionally, TINCR functions as a competing endogenous RNA to upregulate EGFR expression by sponging miR-503-5p.

lnc-MAP3K13-7:1 Inhibits Ovarian GC Proliferation in PCOS via DNMT1 Downregulation-Mediated CDKN1A Promoter Hypomethylation

Xueying Geng et al.

Summary: This study revealed a novel mechanism of DNA hypomethylation induced by lncRNAs, particularly lnc-MAP3K13-7:1, in patients with PCOS, leading to impaired proliferation of GCs. The overexpression of lnc-MAP3K13-7:1 inhibited DNMT1 expression, resulting in increased CDKN1A/p21 expression and cell cycle arrest in the G(0)/G(1) phase. This mechanism acts as a critical pathway in regulating GC growth and may provide potential therapeutic targets for PCOS-related infertility.

A long intergenic non-coding RNA regulates nuclear localization of DNA methyl transferase-1

Rhian Jones et al.

Summary: The nuclear lincRNA CCDC26 plays a crucial role in regulating DNMT1 activity and controlling DNA methylation levels. Its absence leads to mis-localization of DNMT1 to the cytoplasm, causing significant hypomethylation of genomic DNA, double-stranded DNA breaks, and increased cell death. This study reveals a previously unrecognized mechanism of lincRNA-mediated subcellular localization of DNMT1 and regulation of DNA methylation.

A pair of long intergenic non-coding RNA LINC00887 variants act antagonistically to control Carbonic Anhydrase IX transcription upon hypoxia in tongue squamous carcinoma progression

Tao Shen et al.

Summary: This study identified a lncRNA gene related to Chr3q29 in tongue squamous carcinoma and demonstrated how its two variants, 887S and 887L, play opposing roles in tumor progression through the common target CA9. The mechanisms involve the activation of CA9 transcription by 887L through recruiting HIF1 alpha, while 887S suppresses CA9 through DNMT1-mediated DNA methylation. The findings highlight the fine control of hypoxia adaptation through CA9 by these two antagonistic lncRNA variants.

A novel melatonin-regulated lncRNA suppresses TPA-induced oral cancer cell motility through replenishing PRUNE2 expression

Shih-Chi Su et al.

Summary: The inhibitory effect of melatonin on cancer cell dissemination is achieved by downregulating the lncRNA MROS-1, restoring PRUNE2 protein expression, and activating the JAK-STAT pathway. MROS-1, preferentially localized in the nuclei, promotes oral cancer migration by interacting with DNMT3A to modulate PRUNE2 expression. Higher PRUNE2 promoter methylation levels are associated with nodal metastases and decreased PRUNE2 expression in head and neck cancer.
