Advances in the treatment of Chagas disease: Promising new drugs, plants and targets

Paola Garcia-Huertas et al.

Summary: Chagas disease is currently treated with only two drugs, benznidazole and nifurtimox, which have limitations including efficacy in specific infection stages, adverse effects, and parasite resistance. Some institutions and research groups are exploring new therapeutic alternatives through strategies like dosage modification, drug repurposing, combined therapy, the study of medicinal plants, and identification of essential parasite genes for targeted drug design. Collaboration and knowledge exchange among these entities could lead to the development of innovative treatments for Chagas disease.

5-Substituted isatin thiosemicarbazones as inhibitors of tyrosinase: Insights of substituent effects

Run Cheng et al.

Summary: Seven isatin-thiosemicarbazone analogues with different substituents were synthesized and evaluated as inhibitors of mushroom tyrosinase. The substituent modifications affected the interactions between the inhibitors and target molecules, with the thiosemicarbazido moiety playing a predominant role in the inhibition. Theoretical calculations indicated a correlation between the electronic distribution and inhibitory potency of the compounds.

Summary: (English Summary:) This study investigated the potential antichagasic activities of thiosemicarbazones and hydrazones as new anti-T. cruzi drug candidates. Compounds C1 and C3 showed anti-parasitic activity in macrophages without toxicity to host cells, and were also effective in directly killing trypomastigotes. Moreover, C1 and C3 attenuated parasitemia in T. cruzi-infected mice and maintained anti-T. cruzi activity in vivo when loaded in a lipid nanocarrier system.
