Cross Disjoint Mortise Confined Solid-State Nanopores for Single-Molecule Detection

Yexiang Liu et al.

Summary: A concept of cross disjoint mortise confined solid-state nanopore (CDM-Nanopore) fabricated using a focused gallium ion beam and controlled dielectric breakdown technologies is proposed. The confined domain formed by two cross disjoint mortise structures localizes the position for nanopore fabrication with a controlled dielectric breakdown method. Experimental results demonstrate the ion translocation analyses and noise performance of CDM-Nanopore with excellent sensitivities for long-term molecular detection.

Low-Noise Solid-State Nanopore Enhancing Direct Label-Free Analysis for Small Dimensional Assemblies Induced by Specific Molecular Binding

Ruiping Wu et al.

Summary: Solid-state nanopores have shown potential for single-molecular characterization of nucleic acid assemblies and molecular machines. By introducing high-dielectric additives into the test buffer, noise reduction and resolution enhancement can be achieved to directly recognize small dimensional assemblies. This innovation allows for high specificity and improved resolution in distinguishing small molecules without the need for covalent labeling or additional signaling strategies.

DNA Knot Malleability in Single-Digit Nanopores

Rajesh Kumar Sharma et al.

Summary: Researchers found that DNA knots can undergo isomorphic translocation in nanopores, maintaining their equilibrium morphology by rapidly compressing in a lateral direction to fit the constriction. Even for single-digit nanopores, there was no evidence of knot tightening or jamming observed.

Length-Dependent, Single-Molecule Analysis of Short Double-Stranded DNA Fragments through Hydrogel-Filled Nanopores: A Potential Tool for Size Profiling Cell-Free DNA

Dana Al Sulaiman et al.

Summary: Fast sampling and length-dependent detection of short double-stranded DNA fragments in bodily fluids using hydrogel-filled nanopores technology, with a significant increase in signal-to-noise ratio and resolution compared to traditional methods. The technology allows for tunability in both sampling and detection, with increased sampling time leading to higher local DNA concentration at the tip before detection, enabling good resolution of fragments up to 250 bp long.
