Synergistic co-loading of vincristine improved chemotherapeutic potential of pegylated liposomal doxorubicin against triple negative breast cancer and non-small cell lung cancer

Summary: This study explored the biological characteristics of co-loading vincristine with pegylated liposomal doxorubicin in non-small cell lung cancer and triple negative breast cancer. Results showed significantly increased cellular uptake, cell cycle arrest, apoptosis, and reduced cell viability with the co-loaded liposome formulation compared to doxorubicin liposome, leading to improved therapeutic efficacy in vitro and in vivo against NSCLC and TNBC.

Summary: Understanding the factors that influence liposome characteristics such as size, loading efficiency, stability, and drug release can help in the rational design of liposomes for specific pharmacokinetic and biodistribution requirements. This study investigated the effects of formulation methods and parameters on encapsulation efficiency, liposome characteristics, and drug release of PEGylated liposomes containing BIIB093. Various formulation parameters, including lipid chain length, unsaturation, phase transition temperature, and cholesterol content, were found to impact drug loading, release rate, and stability of the liposomes. Encapsulating the drug in the liposome core enabled sustained drug release.

This analytical tool provides added value in determining the optimal drug to lipid ratio in liposomal nano-drugs.
