Summary: This study investigates the structural and thermodynamic aspects of Amphotericin B channel formation, confirming the formation of stable octameric double-length channels and explaining the effects of sterols on channel stability and observed cation selectivity. Through fluorescence lifetime imaging and molecular simulations, the research traces the process of channel assembly and ion permeation, shedding light on the role of tail hydroxyl in the assembly of functional channels and possible origins of cholesterol-ergosterol selectivity.
