Synthesis and Evaluation of a Library of Alternating Amphipathic Copolymers to Solubilize and Study Membrane Proteins

Adrian H. Kopf et al.

Summary: In this study, a library of well-defined alternating copolymers of SMA analogues was designed and synthesized, revealing important polymer properties for membrane protein solubilization and stability. Remarkably, halogenation of the SMA copolymers significantly improved solubilization efficiency, while substituents on the copolymer backbone enhanced resistance to Ca2+.

Interactions of anticancer drugs doxorubicin and idarubicin with lipid monolayers: New insight into the composition, structure and morphology

Dorota Matyszewska et al.

Summary: This study quantifies the interactions of two anthracycline drugs with different lipid monolayers for the first time, showing differences in interaction mechanisms at varying surface pressures. The research provides new insights into the behavior of these drugs in lipid environments, with implications for cancer treatment and drug resistance mechanisms. Advanced techniques, such as neutron reflectometry and Brewster angle microscopy, were used to analyze interfacial composition and structure, revealing important details about drug-lipid interactions.

Understanding the Structural Pathways for Lipid Nanodisc Formation: How Styrene Maleic Acid Copolymers Induce Membrane Fracture and Disc Formation

Victoria Ariel Bjornestad et al.

Summary: Lipid nanodiscs formed by mixtures of styrene maleic acid (SMA) copolymers and lipid membranes are important tools for studying membrane proteins, with the formation pathways depending strongly on factors such as temperature, lipid/ copolymer ratio, and lipid type. The initial aggregation of copolymer in solution leads to insertion of its styrene units into the lipid bilayer hydrocarbon region, causing fractures in the membrane.

A cryo-electron tomography workflow reveals protrusion-mediated shedding on injured plasma membrane

Shrawan Kumar Mageswaran et al.

Summary: The study combines live-cell microscopy, correlative light and electron microscopy, and cryo-ET to investigate shedding in response to plasma membrane damage in HeLa cells, revealing filopodia-like protrusions as scaffolds enriched at damage sites. The shedding process involves F-actin dynamics, myosin-1a, and vacuolar protein sorting 4B, indicating a more complex mechanism than current models of vesiculation. The findings suggest a conserved mechanism with constitutive shedding in enterocytes and the potential for adapting the workflow to study other damage response pathways and dynamic cellular events.
