The Immune Dysregulation of Common Variable Immunodeficiency Disorders

Summary: CVID is a heterogeneous collection of immune dysregulation disorders, with majority of patients experiencing non-infectious complications such as autoimmunity, enteropathy, polyclonal lymphocytosis, and malignancy. Advances in next generation sequencing have led to a better understanding of the genetic basis of CVID disorders, allowing for improved diagnosis and targeted therapies. The identification of cellular abnormalities in T-cell and B-cell compartments, along with the role of innate lymphoid cells in promoting inflammation, provides valuable insights into the immune dysregulation seen in CVID patients.

Summary: 23 cases of thrombotic thrombocytopenia were reported after receiving the AstraZeneca vaccine, incited by antibodies to platelet factor 4 and unrelated to heparin therapy. Avoidance of platelet transfusion and consideration of nonheparin anticoagulant and intravenous immune globulin are recommended for treatment.

Thrombotic Thrombocytopenia after ChAdOx1 nCov-19 Vaccination

Summary: After vaccination with ChAdOx1 nCov-19, rare immune thrombotic thrombocytopenia can occur due to platelet-activating antibodies against PF4, resulting in various thrombotic events. Most patients in this case series were women, with fatal outcomes such as cerebral venous thrombosis and intracranial hemorrhage. Additional studies are needed to further investigate and understand this phenomenon.

Recent studies have identified key distinctions between MDSCs and classical neutrophils and monocytes, providing new insights into therapeutic targeting for cancer and autoimmune diseases through understanding their genomic and metabolic characteristics. Emerging data also suggest potential involvement of MDSCs in pregnancy, neonatal biology, and COVID-19.
