Biosynthesis of ambrein in ambergris: evidence from isotopic data and identification of possible intermediates

Steven J. Rowland et al.

Summary: Ambrein is produced in vivo through enzymatic cyclisation of squalene, exhibiting a different biosynthetic mechanism from co-occurring sterols. Bacterial production of bicyclic polypodenols may be involved in the in vivo biosynthesis of ambrein under certain conditions.

An update on the function and regulation of methylerythritol phosphate and mevalonate pathways and their evolutionary dynamics

Xiaojun Pu et al.

Summary: Isoprenoids are a diverse class of organic compounds involved in photosynthesis, respiration, growth, development, and stress responses in plants. The mevalonate (MVA) and methylerythritol phosphate (MEP) pathways play key roles in isoprenoid synthesis and plant growth, development, and stress responses. The emergence of the MVA pathway is a crucial evolutionary event in plants, facilitating land colonization, embryo development, and adaptation to new environments.

Engineering Saccharomyces cerevisiae for isoprenol production

Jinho Kim et al.

Summary: This study engineered the budding yeast S. cerevisiae for improved biosynthesis of isoprenol, achieving significant yield improvement by employing different biosynthetic pathways and engineering strategies. The implementation of the IPP-bypass pathway and deletion of an endogenous kinase led to a 2-fold increase in isoprenol titer, while metabolomics analysis and overexpression of a promiscuous alkaline phosphatase further boosted the titer to the highest reported level in S. cerevisiae. This work provides key strategies for industrial-scale isoprenol production using yeast as a platform.

RNA sequencing reveals transcriptomic changes in tobacco (Nicotiana tabacum) following NtCPS2 knockdown

Lingxiao He et al.

Summary: This study utilized CRISPR/Cas9 technology to create transgenic tobacco lines with knockdown of NtCPS2 gene, leading to changes in cis-abienol biosynthesis and related compounds in tobacco. RNA sequencing analysis identified numerous differentially expressed genes, highlighting the involvement of various biological and physiological processes such as diterpenoid biosynthesis and plant hormone signal transduction. Overall, this research provides valuable insights into the regulatory mechanisms of cis-abienol biosynthesis in tobacco.
