Fluorovinylsulfones and -Sulfonates as Potent Covalent Reversible Inhibitors of the Trypanosomal Cysteine Protease Rhodesain: Structure-Activity Relationship, Inhibition Mechanism, Metabolism, and In Vivo Studies

Summary: Rhodesain, a cysteine protease of Trypanosoma brucei rhodesiense, has been identified as a drug target for Human African Trypanosomiasis. In this study, alpha-halovinylsulfones were developed as covalent reversible inhibitors for rhodesain based on molecular modeling approaches. The most potent inhibitor 2d showed single-digit nanomolar affinity and selectivity towards mammalian cathepsins B and L, and exhibited slow-tight binding properties. Additionally, the non-fluorinated 2d-(H) demonstrated favorable metabolism and biodistribution in mice brain tissue. Antitrypanosomal activity was highest for inhibitors with specific structural features, with nanomolar EC50 values. The mechanisms of reversible and irreversible inhibitors were elucidated using QM/MM calculations and MD simulations.

Starting from hit compound 1, various spiro scaffolds were generated using a structure-based drug discovery approach, resulting in the development of a promising reversible MAGL inhibitor with significant pharmacodynamic changes in mice.
