Escaping the Lion's Den: redirecting autophagy for unconventional release and spread of viruses

Qi Wen Teo et al.

Summary: Autophagy is an evolutionarily conserved process that maintains cellular homeostasis in response to various stimuli, including unconventional roles such as secretion of cargo without a signal peptide. This nondegradative function of autophagy has been implicated in the infection cycle of various virus species.

ORF3a of the COVID-19 virus SARS-CoV-2 blocks HOPS complex-mediated assembly of the SNARE complex required for autolysosome formation

Guangyan Miao et al.

Summary: ORF3a of the COVID-19 virus SARS-CoV-2 inhibits autophagy activity by blocking fusion of autophagosomes/amphisomes with lysosomes. This leads to accumulation of autophagosomes/amphisomes, impaired lysosome function, and ultimately blocks autophagy process.

Beyond Autophagy: The Expanding Roles of ATG8 Proteins

Jose L. Nieto-Torres et al.

Summary: The ATG8 family proteins are key players in autophagy, participating in cargo recruitment, biogenesis, transport, and fusion with lysosomes. Recent studies have shown their involvement in diverse processes such as vesicle maturation and cargo specification, with complex regulatory mechanisms that are still not fully understood. Individual roles of different ATG8 family members in canonical or non-canonical pathways remain unclear. This review highlights the molecular questions surrounding ATG8s that are only beginning to be explored.

Emerging roles for the autophagy machinery in extracellular vesicle biogenesis and secretion

Andrew M. Leidal et al.

Summary: Autophagy functions to maintain cell health by degrading cytosolic components and recycling them, but it also plays a role in non-degradative processes such as cellular secretion. The coupling of these pathways may have significant impacts on normal physiology and disease.
