Antibody to CD137 Activated by Extracellular Adenosine Triphosphate Is Tumor Selective and Broadly Effective In Vivo without Systemic Immune Activation

Mika Kamata-Sakurai et al.

Summary: The novel anti-CD137 switch antibody STA551 shows strong and broad antitumor efficacy by targeting extracellular ATP in the tumor microenvironment, providing a potential treatment option for a wide variety of cancers regardless of antigen expression. The antibody demonstrates potent tumor inhibition without systemic toxicity or dependence on antigen expression in both mouse and human tumor models, supporting its further clinical testing and potential application to other cancer therapy targets.

Trastuzumab-induced cardiotoxicity: a review of clinical risk factors, pharmacologic prevention, and cardiotoxicity of other HER2-directed therapies

Naomi Dempsey et al.

Summary: While trastuzumab has been successful in treating breast cancer, it may lead to heart failure and loss of left ventricular function. Risk factors for trastuzumab-induced cardiotoxicity include cardiovascular disease, older age, and prior anthracycline treatment, with anthracycline exposure being the most significant. Cardiac biomarkers and imaging assessments can help in early identification of cardiotoxicity, and initiation of certain medications may prevent this complication. Other HER2-targeted therapies seem to have lower rates of cardiotoxicity compared to trastuzumab, with anthracycline-free regimens now considered standard of care for early HER2-positive breast cancer.

A general Fc engineering platform for the next generation of antibody therapeutics

Da Chen et al.

Summary: Fc engineering has become a focus in antibody drug development. A mammalian cell display-based platform was developed to screen millions of Fc variants for optimized binding to Fc gamma RIIIa or Fc gamma RIIb. Novel Fc variants with enhanced binding and improved effector function were successfully identified.
