Emerging Concepts of Endotypes/Phenotypes in Regenerative Medicine for Osteoarthritis

Gun-Il Im et al.

Summary: Given the lack of consistent success in structural improvement with regenerative medicine for OA, introducing the concept of phenotype/endotype can help identify suitable patients for costly treatments, potentially leading to better treatment outcomes. A better understanding of molecular endotypes can help define clinical phenotypes more clearly and guide treatment decisions.

Cartilage Targets of Knee Osteoarthritis Shared by Both Genders

Chenshuang Li et al.

Summary: OA affects people of all ages, sexes, and races, with a greater burden in women. Gender-specific OA management shows potential, and drug development should consider OA-responsive biomarkers. Genes from a transcriptome dataset are analyzed to identify key pathways influencing OA.

Disease-modifying therapeutic strategies in osteoarthritis: current status and future directions

Yongsik Cho et al.

Summary: OA is the most common form of arthritis characterized by cartilage destruction and chronic pain. Current treatments aim to relieve symptoms, but new approaches such as DMOADs and regenerative methods targeting inflammatory cytokines and stimulating cartilage regeneration offer promise in controlling or reversing the disease. Researchers are exploring these avenues based on molecular understanding and some approaches are nearing general clinical use.

Current Models for Development of Disease-Modifying Osteoarthritis Drugs

Meagan J. Makarczyk et al.

Summary: Currently, there are no approved disease-modifying osteoarthritis drugs (DMOADs) for widespread clinical use. The lack of effective DMOADs may be due to various factors, including the differences between human knee joints and preclinical models. This review discusses the current OA models, highlights the need for future model development, and emphasizes the potential of microphysiological systems in drug development.

Males and Females Have Distinct Molecular Events in the Articular Cartilage during Knee Osteoarthritis

Chenshuang Li et al.

Summary: There are significant differences at the transcriptomic level in knee articular cartilage among healthy and OA individuals, particularly with regards to sex distinctions. Females showed more differential gene expression and enriched pathways in comparison to males, both in healthy cartilage and in OA conditions. Male and female cartilage responded differently to OA, with notable differences in the expression of genes related to various pathways, suggesting potential implications for OA prognosis and treatment outcomes in males and females.
