Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Acute Complications and Survival Analysis of Childhood Acute Lymphoblastic Leukemia: A 15-year Experience

Ayse Pinar Ozturk et al.

Summary: The study evaluated the acute complications and survival rates of childhood acute lymphoblastic leukemia in 110 patients treated with the Children's Oncology Group protocol. The most common complication was infection, followed by gastrointestinal, neurologic, metabolic/endocrine, drug-related hypersensitivity, avascular necrosis, thrombotic, severe psychiatric, and various other complications. The 5- and 10-year overall survival rates were both 85.9%, with relapse-free survival rates at 1, 3, and 5 years being 97.9%, 91.3%, and 86.3%, respectively.

Summary: The epidemiologic progress against childhood and adolescent acute lymphoblastic leukemia in the Netherlands showed stable overall incidence rates, despite increases in certain subtypes in different age groups. The survival rates for patients have improved, while the mortality rates have decreased, primarily due to treatment in pediatric oncology centers.

TLR9 expression in chronic lymphocytic leukemia identifies a promigratory subpopulation and novel therapeutic target

Emma Kennedy et al.

Summary: Despite the advancements in B-cell receptor-targeted inhibitors, CLL remains incurable. TLR9 activation by unmethylated DNA is associated with disease progression in CLL. High TLR9 expression promotes CLL cell migration and disease progression, while dual targeting of TLR9 and BTK shows strong synergism as a potential treatment strategy for this incurable disease.

Summary: MicroRNAs play an active role in cancer development, and restoring the expression of tumor suppressor miRNAs can be a viable treatment option. Combining chemotherapy with miRNAs can help overcome drug resistance and improve the efficacy of cancer treatment.

DNA methylation-mediated silencing of microRNA-204 enhances T cell acute lymphoblastic leukemia by up-regulating MMP-2 and MMP-9 via NF-κB

Congmeng Lin et al.

Summary: T cell acute lymphoblastic leukaemia (T-ALL) is characterized by abnormal expression of microRNAs (miRNAs), with miR-204 down-regulation in T cells of T-ALL patients due to increased DNA methylation. Overexpression of miR-204 inhibits T-ALL cell proliferation and enhances apoptosis through interleukin receptor-associated kinase 1 (IRAK1).

MiR-454-3p Promotes Oxaliplatin Resistance by Targeting PTEN in Colorectal Cancer

Xiao-Lan Qian et al.

Summary: The study identified that miR-454-3p is up-regulated in oxaliplatin-resistant colorectal cancer cells, promoting resistance by targeting PTEN and activating the AKT pathway. Inhibition of miR-454-3p restored sensitivity to oxaliplatin in resistant cells and enhanced apoptosis. Furthermore, overexpression of miR-454-3p correlated with decreased responsiveness to oxaliplatin-based chemotherapy and shorter progression-free survival.

Targeting Autophagy Triggers Apoptosis and Complements the Action of Venetoclax in Chronic Lymphocytic Leukemia Cells

Damjan Avsec et al.

Summary: Continuous treatment with Venetoclax can lead to resistance in patients with CLL, highlighting the need for novel targets. Autophagy inhibitors show potential as effective targets, exhibiting cytotoxic activities against CLL cells and complementing the action of Venetoclax. These inhibitors demonstrate selectivity towards CLL cells and have synergistic effects when combined with Venetoclax, providing a rationale for targeting autophagy in CLL treatment.
