Anticancer Activity of the Acetylenic Derivative of Betulin Phosphate Involves Induction of Necrotic-Like Death in Breast Cancer Cells In Vitro

Summary: In this study, it was found that 30-diethoxyphosphoryl-28-propynoylbetulin displayed the strongest inhibitory effect on growth and induction of cellular death in human breast cancer cells. The compound decreased DNA synthesis, up-regulated p21(WAF1/Cip1) levels, increased cell death, and triggered reactive oxygen species generation and mitochondrial membrane potential loss, ultimately leading to a form of regulated cell demise that exhibits features of necrosis-like morphology and activation of caspases.

Molecular Structure, In Vitro Anticancer Study and Molecular Docking of New Phosphate Derivatives of Betulin

Summary: A series of 30-diethylphosphate derivatives of betulin were synthesized and evaluated for their cytotoxic activity against human cancer cell lines. Compounds 7a and 7b showed the highest activity against C-32 and SNB-19 cell lines, with IC50 values of 2.15 and 0.91 mu M for 7a, and 0.76 and 0.8 mu M for 7b. The most active compounds were further studied for their effects on apoptosis markers and docked to the active site of the EGFR protein, showing significant interactions and stable complexes.

Effect of F16-Betulin Conjugate on Mitochondrial Membranes and Its Role in Cell Death Initiation

Summary: This study demonstrates the targeted effects of a newly synthesized F16-betulin conjugate on mitochondria and its cytotoxicity. The conjugate reduces mitochondrial potential, induces superoxide overproduction, and surface-active aggregation of mitochondria. It also suppresses respiration, oxidative phosphorylation, and increases H2O2 generation, possibly through inhibition of complex I of the mitochondrial electron transport chain.

High cytotoxicity of betulin towards fish and murine fibroblasts: Is betulin safe for nonneoplastic cells?
