RS_CRZ1, a C2H2-Type Transcription Factor Is Required for Pathogenesis of Rhizoctonia solani AG1-IA in Tomato

Srayan Ghosh et al.

Summary: The study successfully employed HIGS and dsRNA technologies to downregulate pathogenic genes of R. solani in tomato, with significant reduction in disease symptoms and pathogen colonization depth observed upon silencing of the RS_CRZ1 gene. Additionally, coregulated genes with RS_CRZ1 were identified during the pathogenicity process, highlighting the importance of RS_CRZ1 in managing the hostile environment encountered during host colonization and the potential of gene silencing approaches for functional characterization of pathogenicity determinants of R. solani.

The C2H2 Transcription Factor SltA Contributes to Azole Resistance by Coregulating the Expression of the Drug Target Erg11A and the Drug Efflux Pump Mdr1 in Aspergillus fumigatus

Wenlong Du et al.

Summary: The discovery of the new C2H2 zinc finger transcription factor SltA in regulating azole resistance marks a milestone. SltA not only participates in its own transcriptional regulation, but also directly binds to the conserved sequences in the promoter regions of ergosterol biosynthesis and drug efflux pump related genes. Deletion of sltA results in hypersusceptibility to itraconazole. The absence of sltA significantly reduces drug resistance in laboratory-derived and clinical azole-resistant isolates.

Specific interaction of an RNA-binding protein with the 3′-UTR of its target mRNA is critical to oomycete sexual reproduction

Hui Feng et al.

Summary: This study introduces a CRISPR/Cas9 system for gene manipulation in the oomycete pathogen Pythium ultimum, revealing the regulatory role of the RNA-binding protein PuM90 in oospore formation. The specific interaction between PuM90 and the 3'-UTR of PuFLP mRNA is critical for sexual reproduction in P. ultimum.

A Cytochrome B5-Like Heme/Steroid Binding Domain Protein, PlCB5L1, Regulates Mycelial Growth, Pathogenicity and Oxidative Stress Tolerance in Peronophythora litchii

Wen Li et al.

Summary: This study identified a gene encoding a Cyt-b(5) domain protein in the plant pathogenic oomycete Peronophythora litchii, highlighting its crucial role in development, stress response, and pathogenicity. Knockout mutants of PlCB5L1 showed reduced growth rate, altered stress tolerance, and impaired pathogenicity, demonstrating the significance of Cyt-b(5) domain proteins in plant pathogenic oomycetes.

The Mitogen-Activated Protein Kinase PlMAPK2 Is Involved in Zoosporogenesis and Pathogenicity of Peronophythora litchii

Jiamin Huang et al.

Summary: This study identified a critical MAPK, PlMAPK2, in the plant pathogenic oomycete Peronophythora litchii, which plays a key role in zoosporogenesis. PlMAPK2 mutants showed impaired sporangium release, reduced laccase activity, and decreased virulence to lychee leaves.
