Fentanyl Structure as a Scaffold for Opioid/Non-Opioid Multitarget Analgesics

Synthesis and evaluation of peptide-fentanyl analogue conjugates as dual μ/δ-opioid receptor agonists for the treatment of pain

Summary: Novel peptide-fentanyl analogue conjugates were synthesized by coupling carfentanyl derivatives to the conformationally constrained dermorphin tetrapeptide BVD03. These conjugates exhibited distinct binding and functional activities at opioid receptors and demonstrated analgesic effects when conjugated to the peptide. The most potent compound, SW-LJ-11, displayed mixed agonist properties at opioid receptors and showed significant analgesic efficacy in classic mouse pain models. Importantly, SW-LJ-11 did not induce physical dependence or respiratory depression, suggesting that opioid peptide-fentanyl analogue conjugates could be a promising strategy for developing safer opioids.
