Proteomic Analysis of Methylglyoxal Modifications Reveals Susceptibility of Glycolytic Enzymes to Dicarbonyl Stress

Leigh Donnellan et al.

Summary: In this study, over 500 MGO modified proteins were identified through proteomic analysis, with a notable over-representation of modifications on glycolytic enzymes, ribosomal, and spliceosome proteins. Furthermore, MGO modifications were found on the active site residues of glycolytic enzymes, potentially affecting their activity. The modification of fructose bisphosphate aldolase was consistently observed in various cell lines and peripheral blood lymphocytes.

Dicarbonyl stress, protein glycation and the unfolded protein response

Naila Rabbani et al.

Summary: Research suggests that dicarbonyl stress may be an important physiological stimulus for insulin resistance and inducers of Glo1 could provide an effective route for suppression and therapy. Further investigation and validation may provide new insights into physiological activators of the UPR and their association with dicarbonyl stress.

Non-enzymatic Covalent Modifications as a New Chapter in the Histone Code

Igor Maksimovic et al.

Summary: The interior of the cell contains reactive species that can accumulate as non-enzymatic covalent modifications (NECMs) on biological macromolecules, interfering with cellular processes and affecting chromatin structure and transcriptional output. Known cellular regulatory mechanisms can prevent or reverse the accumulation of these covalent modifications.

Causes and consequences of micronuclei

Ksenia Krupina et al.

Summary: Micronuclei are small compartments with DNA content encapsulated by a nuclear envelope, found in cancers, during senescence, and after genotoxic stress. Compromised integrity of the micronuclear envelope disrupts DNA replication and repair, exposing micronuclear DNA to cytoplasm. Micronuclei play a central role in tumorigenesis, being a source of complex genome rearrangements and promoting a cellular immune response that may contribute to cancer metastasis.

Methylglyoxal induces chromosomal instability and mitotic dysfunction in lymphocytes

Leigh Donnellan et al.

Summary: Type 2 diabetes is associated with elevated levels of DNA damage, particularly micronuclei, caused by reactive dicarbonyl MGO. MGO exposure leads to increased chromosomal instability and DNA damage, affecting genomic integrity.
