Summary: This study describes the first-in-man study of a replication-deficient arenavirus-based vector, HB-101, expressing cytomegalovirus gB and pp65. Consecutive administrations of HB-101 were well tolerated and elicited gB-binding and neutralizing antibodies as well as cytomegalovirus-specific CD8 T-cell responses in healthy volunteers.

Heterologous arenavirus vector prime-boost overrules self-tolerance for efficient tumor-specific CD8 T cell attack

Summary: This study engineered two distantly related arenaviruses for therapeutic cancer vaccination and demonstrated that a prime-boost regimen with these viruses induced tumor-specific CTL responses and eliminated established solid tumors in mice. The findings suggest that combining two genealogically distantly related arenaviruses allows reshuffling of immunodominance hierarchies and breaking self-directed tolerance for effective tumor control.
