Significance of CD47 and Its Association With Tumor Immune Microenvironment Heterogeneity in Ovarian Cancer

Lan Yu et al.

Summary: This study demonstrated the upregulation of CD47 in ovarian cancer and its correlation with poor overall survival and progression-free survival. CD47 expression levels were closely related to immune infiltration in ovarian cancer, particularly with Treg cells, monocytes, macrophages, and T cell exhaustion. The distinct cell components and distribution in primary and metastatic ovarian cancer may contribute to the tumor microenvironment heterogeneity.

LncRNA TCF7 Promotes Epithelial Ovarian Cancer Viability, Mobility and Stemness via Regulating ITGB8

Changlei Su et al.

Summary: The study revealed that lnc-TCF7 regulates multiple oncogenic pathways in epithelial ovarian cancer by upregulating ITGB8, promoting cell proliferation, migration, invasion, and stemness. Additionally, high expression of lnc-TCF7 is associated with higher pathological grade, larger tumor size, advanced FIGO stage, and poorer overall survival in EOC patients.

Atezolizumab, Bevacizumab, and Chemotherapy for Newly Diagnosed Stage III or IV Ovarian Cancer: Placebo-Controlled Randomized Phase III Trial (IMagyn050/GOG 3015/ENGOT-OV39)

Kathleen N. Moore et al.

Summary: The study evaluated the addition of the humanized monoclonal antiprogrammed death ligand-1 (PD-L1) antibody atezolizumab to platinum-based chemotherapy and bevacizumab in newly diagnosed stage III or IV ovarian cancer, showing a significant progression-free survival advantage in the PD-L1-positive population. However, no significant overall survival benefit was observed from atezolizumab in the interim analysis. The most common grade 3 or 4 adverse events were neutropenia, hypertension, and anemia.

NK Cell-Mediated Eradication of Ovarian Cancer Cells with a Novel Chimeric Antigen Receptor Directed against CD44

Ruediger Klapdor et al.

Summary: Ovarian cancer stem cells with chemoresistance contribute to tumor recurrence, leading to poor survival rates. Targeting CD44 using CAR immunotherapy demonstrates potent cytotoxic activity against CD44-positive ovarian cancer cells. Furthermore, combined treatment of CD44-targeted therapy and cisplatin shows higher anti-tumor activity compared to sequential treatment.

Targeting Ovarian Cancer Stem Cells by Dual Inhibition of HOTAIR and DNA Methylation

Weini Wang et al.

Summary: The study demonstrated that the overexpression of long noncoding RNA HOTAIR in ovarian high-grade serous carcinoma cell lines promotes cancer stem cells and chemoresistant tumors. By disrupting the interaction between HOTAIR and EZH2 with a peptide nucleic acid, combined with a DNMT inhibitor, the formation of OCSCs was inhibited, providing a potential approach to eradicate OCSCs and prevent disease recurrence.

LINC00115 promotes stemness and inhibits apoptosis of ovarian cancer stem cells by upregulating SOX9 and inhibiting the Wnt/β-catenin pathway through competitively binding to microRNA-30a

Rui Hou et al.

Summary: In ovarian cancer cells, LINC00115 and SOX9 were found to be highly expressed, while miR-30a was lowly expressed. The study demonstrated that LINC00115 promotes stemness and inhibits apoptosis of ovarian cancer stem cells by upregulating SOX9 and activating the Wnt/beta-catenin pathway through competitively binding to miR-30a. Bioinformatics analysis and experimental validation also revealed that miR-30a can target SOX9, while LINC00115 can bind to miR-30a.

Summary: Retrospective studies have shown the prognostic value of intra-tumoral immune cell infiltration density in OC, but clinical trials evaluating PD1/PD-L1 inhibitors in relapsed OC patients have been disappointing. Strategies to enhance the benefit from immune-oncology agents in OC include combining anti-PD-L1 with other drugs, evaluating these agents earlier in the disease course, or in biomarker selected patients.
