Melanocytes as emerging key players in niche regulation of limbal epithelial stem cells

Naresh Polisetti et al.

Summary: Limbal melanocytes play crucial roles in the corneal epithelial stem cell niche, protecting stem cells from UV damage and regulating immune responses and angiogenesis. Their diverse functions beyond melanin production have significant implications for regenerative cell therapies in the future.

Process development and safety evaluation of ABCB5+ limbal stem cells as advanced-therapy medicinal product to treat limbal stem cell deficiency

Alexandra Norrick et al.

Summary: This study developed a novel treatment strategy utilizing in vitro expanded allogeneic ABCB5(+) LSCs derived from human cadaveric limbal tissue. The product showed high percentages of cells expressing critical transcription factors for LSC stemness maintenance and corneal epithelial differentiation. Preclinical studies confirmed the engraftment potential of the cells and demonstrated their safety, leading to approval for a clinical trial to evaluate safety and therapeutic efficacy in patients with LSCD.
