Neutrophil-Platelet Interactions as Novel Treatment Targets in Cardiovascular Disease

Rainer Kaiser et al.

Summary: Neutrophils and platelets, two abundant cell types in peripheral blood, play important roles in immune responses and clot formation in cardiovascular diseases. This review describes their physical interactions, the impact of release of granule contents on each other, and their contribution to thromboinflammation in COVID-19. Targeting neutrophil-platelet interactions may provide a novel treatment strategy in cardiovascular disease.

Autoantibody-mediated desialylation impairs human thrombopoiesis and platelet lifespan

Irene Marini et al.

Summary: In this study, novel effector functions of autoantibodies in patients with immune thrombocytopenia were identified, which interfere with the clinical presentation of the disease by inducing cleavage of sialic acid residues from the surface of platelets and megakaryocytes. Additionally, desialylation mediated by autoantibodies disrupts the interaction between cells and extracellular matrix proteins, leading to impaired platelet adhesion and megakaryocyte differentiation. This suggests that prevention of desialylation may be a potential therapeutic approach in the future clinical studies for treating bleeding in immune thrombocytopenia.

Desialylation of O-glycans on glycoprotein Ibα drives receptor signaling and platelet clearance

Summary: During infection, neuraminidase removes sialic acid from platelet GPIb alpha, leading to their rapid clearance. This mechanism may explain many puzzling and seemingly contradictory observations associated with clearance of desialylated or hyposialylated platelets.

Differential regulation of the platelet GPIb-IX complex by anti-GPIbβ antibodies

M. Edward Quach et al.

Summary: The study identified a novel rat monoclonal antibody 3G6 that acts as a positive modulator of GPIb beta, enhancing GPIb-IX binding and activation. In contrast, another antibody RAM.1 inhibits GPIb-IX signaling, suggesting that changes in GPIb beta conformation may be crucial for outside-in activation via GPIb-IX.

Circulating platelet count and glycans

Leonardo Rivadeneyra et al.

Summary: This review highlights the crucial role of carbohydrates, particularly sialic acid moieties, in mediating platelet clearance and controlling platelet numbers. The production rate by megakaryocytes and removal kinetics are key factors in regulating circulating platelet count, with alterations potentially leading to thrombocytopenia or thrombocytosis. Understanding carbohydrate-related mechanisms is essential for maintaining a steady-state platelet count and preventing adverse events.

Leukocyte trafficking to the lungs and beyond: lessons from influenza for COVID-19

Summary: The understanding of COVID-19 clinical manifestations requires knowledge of how immune cells are recruited to virus-infected lungs and how this recruitment varies among individuals. The orchestration of leukocyte trafficking to respiratory system is controlled by specific molecules, which can lead to pathological complications when uncontrolled. The incomplete understanding of trafficking signals in response to SARS-CoV-2 infection calls for further research for potential therapeutic interventions.
