Strategies to prolong the residence time of drug delivery systems on ocular surface

Brunella Grassiri et al.

Summary: Ocular diseases can be treated via various routes of administration, with topical administration being the most accepted by patients despite its lower bioavailability. To prolong drug residence time and reduce dosing frequency, strategies like viscosity increasing and mucoadhesive excipients are used. Liquid and semisolid formulations are preferred over solid ones for their potential to contain nano- and microcarrier systems that contribute to prolonged residence time.

Thiolated cyclodextrins: Mucoadhesive and permeation enhancing excipients for ocular drug delivery

Mulazim Hussain Asim et al.

Summary: Thiolated beta-cyclodextrin was synthesized and characterized for its potential use as an auxiliary agent for ocular drug delivery, showing enhanced permeation and mucoadhesive properties. The newly synthesized thiolated oligomer exhibited improved aqueous solubility, viscosity enhancement of mucus, prolonged ocular residence time, and enhanced permeation of sodium fluorescein on various ocular tissues, making it a promising candidate for ocular drug delivery.

Spray-dried mucoadhesive microparticles based on S-protected thiolated hydroxypropyl-β-cyclodextrin for budesonide nasal delivery

Giuseppe Francesco Racaniello et al.

Summary: This study demonstrated that thiolated hydroxypropyl-beta-cyclodextrin can improve the water solubility of Budesonide and be used to prepare mucoadhesive microparticles for intranasal delivery. The spray-dried microparticles showed good stability, rapid dissolution, and prolonged mucosal residence time, making them a promising tool for the nasal delivery of poorly water-soluble corticosteroids.
