Fabrication and biomedical applications of Arabinoxylan, Pectin, Chitosan, soy protein, and silk fibroin hydrogels via laccase-Ferulic acid redox chemistry

Lignin-oxidizing activity of bacterial laccases characterized using soluble substrates and polymeric lignin

Summary: Efficient lignin biotransformation requires the activity of different oxidative enzymes. This study identified the bacterial multi-copper oxidase laccase CotA(Bsu) from Bacillus subtilis as having the highest activity against soluble substrates. Site-directed mutagenesis of CotA(Bsu) revealed critical residues for activity and a lignin-agarose plate screen confirmed the results, showing increased activity towards kraft lignin for certain mutant variants. X-ray photoelectron spectroscopy analysis of low sulfonate kraft lignin after incubation with CotA(Bsu) indicated a reduction in C-C/C=C bonds and an increase in C-O/C=O bonds. Product analyses suggested radical intermediates followed by radical coupling in the oxidation of lignin by laccases.

Substrate specificity of a new laccase from Trametes polyzona WRF03

Summary: In this study, various aromatic compounds structurally similar to lignin were tested as substrates for purified laccase from Trametes polyzona WRF03, with ABTS showing the highest specificity. The oxidation of phenolic substrates was found to be influenced by the nature and position of substituent groups on the aromatic nucleus. These findings suggest potential applications of the enzyme in delignification and treatment of wastewater containing dyes and PAHs.
