Neutrophil-derived extracellular vesicles promote feed-forward inflammasome signaling in cystic fibrosis airways

Osric A. Forrest et al.

Summary: Neutrophils in cystic fibrosis (CF) airways contribute to chronic inflammation by packaging proinflammatory signals in extracellular vesicles (EVs). These EVs can induce reprogramming of naive neutrophils and activate caspase-1 and IL-1β production. Additionally, EVs from activated neutrophils can deliver active caspase-1 to tracheal epithelial cells and induce the release of IL-1α.

Changes in airway inflammation with pseudomonas eradication in early cystic fibrosis

Luke W. Garratt et al.

Summary: This study evaluated the changes in neutrophil elastase (NE) activity following Pseudomonas aeruginosa eradication in children with cystic fibrosis. The results showed that NE activity was significantly reduced post-eradication, but persisted in some subjects. High post-eradication NE levels were associated with an increased risk for P. aeruginosa infection in the next annual visit.

Transcriptional firing represses bactericidal activity in cystic fibrosis airway neutrophils

Camilla Margaroli et al.

Summary: The study reveals that neutrophils recruited to the airways of cystic fibrosis patients exhibit rapid and broad transcriptional activity, leading to upregulation of anabolic genes and downregulation of antimicrobial genes. Inhibition of RNA polymerase II and III restores the expression of key antimicrobial genes, increasing the bactericidal capacity of neutrophils.
