GSDMB is increased in IBD and regulates epithelial restitution/repair independent of pyroptosis

Nitish Rana et al.

Summary: Increased GSDMB in inflammatory bowel disease affects the restoration of epithelial barrier function and resolution of inflammation. Mechanistic experiments reveal that GSDMB deficiency impairs the increased proliferation and migration of epithelial cells during in vitro wound closure.

Tuning tuft cells: new ligands and effector functions reveal tissue-specific function

Tyler E. Billipp et al.

Summary: Tuft cells are rare chemosensory epithelial cells that monitor the environment and initiate immune responses in different tissues based on their unique receptors and effector functions tuned to the surrounding tissue's physiology.

The metabolite α-KG induces GSDMC-dependent pyroptosis through death receptor 6-activated caspase-8

Jia-yuan Zhang et al.

Summary: Pyroptosis is induced by alpha-ketoglutarate through caspase-8-mediated cleavage of GSDMC. This process involves elevation of ROS levels leading to oxidation of DR6 and endocytosis, ultimately resulting in pyroptosis. The acidic environment and production of L-2HG play a crucial role in enhancing the efficiency of alpha-KG-induced pyroptosis in cancer cells.

Up-regulation of gasdermin C in mouse small intestine is associated with lytic cell death in enterocytes in worm-induced type 2 immunity

Ranhui Xi et al.

Summary: Taste-like tuft cells in the intestine trigger type 2 immunity in response to worm infection, leading to the clearance of worms from the gut via type 2 immune responses. The up-regulated gasdermin C genes (Gsdmcs) may be major effectors for type 2 responses in the gut, and Gsdmc-mediated pyroptosis may provide a conduit for the release of antiparasitic factors from enterocytes to facilitate the clearance of worms.

Spatial discordances between mRNAs and proteins in the intestinal epithelium

Yotam Harnik et al.

Summary: The study identified spatial discrepancies between enterocyte mRNA and protein levels in the intestinal epithelium, providing a spatial blueprint of proteins along the villi. Through spatial sorting and Bayesian methods, it was found that while many genes exhibit zonated proteins towards the villus tip, mRNA is zonated towards the villus bottom. Independent protein synthesis delays in space were shown to explain many mRNA-protein discordances.
