Therapeutic potential of melatonin and melatonergic drugs on K18-hACE2 mice infected with SARS-CoV-2

Erika Cecon et al.

Summary: As the COVID-19 pandemic continues to spread, various therapeutic candidates are being tested, but there is no definitive treatment for those already infected. Melatonin and its compounds have shown promise in delaying severe clinical outcomes in a mouse model and improving survival rates, especially at high doses. Additionally, these compounds also limit the exaggerated production of interferons in the lungs, potentially contributing to improved symptoms in treated mice. Further studies are needed to explore the potential benefits of repurposing melatonergic drugs for treating COVID-19 and related diseases in humans.

Melatonin in the Prophylaxis of SARS-CoV-2 Infection in Healthcare Workers (MeCOVID): A Randomised Clinical Trial

Irene Garcia-Garcia et al.

Summary: In this randomized, double-blind clinical trial, we evaluated the efficacy of melatonin as a prophylactic treatment for preventing SARS-CoV-2 infection among healthcare workers at high risk of exposure. The results showed that the incidence of SARS-CoV-2 infection was 5.5% in the melatonin group and 2.6% in the placebo group, indicating no significant difference. However, the melatonin group experienced more treatment-related adverse events compared to the placebo group.

The prevalence of depression, anxiety, and sleep disturbances in COVID-19 patients: a meta-analysis

Summary: Evidence from previous coronavirus outbreaks indicates that COVID-19 patients are at risk of developing psychiatric disorders such as depression, anxiety, and sleep disturbances. A systematic review and meta-analysis found high prevalence rates of depression (45%), anxiety (47%), and sleep disturbances (34%) in COVID-19 patients. Prevalence estimates varied based on screening tools used, highlighting the need for more studies on mental well-being in COVID-19 patients.

Summary: This study investigated the prevalence of delirium and coma in critically ill patients with COVID-19, identifying benzodiazepine use and lack of family visitation as modifiable risk factors for delirium. These findings present an opportunity to reduce acute brain dysfunction in patients with COVID-19.

Summary: The study found that COVID-19 pneumonia patients treated with prolonged-release melatonin 2 mg had a shorter duration of non-invasive ventilation, shorter stay in sub-intensive care, and lower incidence of delirium compared to untreated patients. These results suggest potential benefits of prolonged-release melatonin 2 mg therapy in COVID-19 treatment.
