Competition between two phosphatases fine-tunes Hedgehog signaling

Min Liu et al.

Summary: Protein phosphatase V (PpV) is identified as a homeostatic regulator of Hh signaling, which competes with the catalytic subunit of PP2A for Wdb association to negatively regulate Wdb stability. This competition ensures graded Hh signaling through regulated Wdb stability. PpV functions as a Hh activity sensor to maintain Hh signaling homeostasis by regulating Wdb-mediated PP2A activity through feedback mechanisms.

Tubulin glycylation controls axonemal dynein activity, flagellar beat, and male fertility

Sudarshan Gadadhar et al.

Summary: This study demonstrates the importance of tubulin glycylation for controlled flagellar beating, directional sperm swimming, and male fertility by utilizing a mouse model lacking this modification. The lack of glycylation led to abnormal dynein arm conformations in sperm axonemes, resulting in subfertility in male mice due to aberrant beat patterns of sperm flagella.

The Emerging Roles of Axonemal Glutamylation in Regulation of Cilia Architecture and Functions

Wen-Ting Yang et al.

Summary: Cilia, highly conserved hair-like structures protruding from cell surface among diverse species, play crucial roles in cell functions. Axonemal glutamylation, a post-translational modification in ciliary axonemes, regulates the structure and function of cilia. Investigating the physiological roles of axonemal glutamylation helps in understanding its significance in human health.

Ciliary extracellular vesicles are distinct from the cytosolic extracellular vesicles

Ashraf M. Mohieldin et al.

Summary: Extracellular vesicles (EVs) are cell-derived membrane vesicles that mediate intercellular signalling pathways. Primary cilia, mechanosensory organelles, are associated with human diseases. This study found distinct protein profiles between ciliary EVs and cytosolic EVs, highlighting unique differences in vesicles secreted from cilia and the cytosol.

Phosphorylation and Ubiquitylation Regulate Protein Trafficking, Signaling, and the Biogenesis of Primary Cilia

Elena A. May et al.

Summary: The primary cilium is a microtubule-based membrane protrusion that senses the cellular environment and triggers specific cellular responses, requiring diverse components and regulated interplay. Recent research has focused on cilia-specific phosphorylation and ubiquitylation events, shedding light on the molecular mechanisms that regulate cilia functions and their implications for biogenesis, protein trafficking, and signaling processes.
