The Mineralization Regulator ANKH Mediates Cellular Efflux of ATP, Not Pyrophosphate

Summary: The plasma membrane protein ANKH regulates pathological mineralization of joints by controlling extracellular levels of the mineralization inhibitor PPi. Recent research shows that ANKH overexpression results in the release of large amounts of NTPs, primarily ATP, into the culture medium. This ATP is then converted into PPi and AMP by the enzyme ENPP1. The study suggests that ANKH may not only transport PPi but also release it directly.

Inhibition of the DNA Damage Response Attenuates Ectopic Calcification in Pseudoxanthoma Elasticum

Summary: The DNA damage response (DDR) and poly(ADP-ribose) (PAR) pathways are involved in the ectopic calcification in pseudoxanthoma elasticum (PXE). Inhibition of PAR polymerase 1 (PARP1) and minocycline treatment can reduce calcification. These findings suggest that local DDR/PAR signaling contributes to the pathogenesis of PXE and its inhibition may be a promising treatment strategy for ectopic calcification in PXE.

Summary: The study found that plasma PPi levels are significantly reduced in SSc patients with or without calcinosis, suggesting a potential pathogenic role in calcinosis cutis.
