Spray-Congealing and Wet-Sieving as Alternative Processes for Engineering of Inhalation Carrier Particles: Comparison of Surface Properties, Blending and In Vitro Performance

Joana T. Pinto et al.

Summary: This study explored various particle engineering processes to produce D-mannitol carriers for inhaled delivery. Wet-sieving produced D-mannitol particles with narrow size distribution, while spray-congealing generated free-flowing spherical particles. Wet-sieved D-mannitol particles showed benefits for drug aerosolization, especially when combined with a ternary agent, while spray-congealed D-mannitol exhibited promising results in increasing the fine particle fraction of the drug without additional agents.
