Recent Developments in the Synthesis of Tetrazoles and their Pharmacological Relevance

Socorro Leyva-Ramos et al.

Summary: The heterocycle ring tetrazole is an important structural moiety in medicinal chemistry, acting as a bioisosteric analogue of carboxylic acid and cis-amide groups. It exhibits a wide range of biological activities, including antiviral, antibacterial, anticancer, antifungal, and antioxidant properties. Various synthetic methods, such as using azide derivatives and different heterogeneous catalysts, have been developed to synthesize tetrazole derivatives.

A One-Pot Six-Component Reaction for the Synthesis of 1,5-Disubstituted Tetrazol-1,2,3-Triazole Hybrids and Their Cytotoxic Activity against the MCF-7 Cell Line

Cesia M. Aguilar-Morales et al.

Summary: This study presents a high-order multicomponent reaction to obtain novel linked 1,5-disubstituted tetrazole-1,2,3-triazole hybrids in low to moderate yield. The cascade process involves Ugi-azide, bimolecular nucleophilic substitution (S(N)2), and copper-catalyzed alkyne-azide reaction (CuAAC), forming six new bonds. The synthesized hybrid compounds show potential as drug candidates for breast cancer treatment, especially those with cyclohexane and halogens in their structures, although further assays are needed to fully understand their effects on cancer cell hallmarks.

Synthesis of phthalimides, isoindolin-1-ones and isoindolines bearing aminobenzoic acids as a new fluorescent compounds

Melchor Solis-Santos et al.

Summary: Both experimental and theoretical methods were used to study the fluorescent properties of nine new compounds based on phthalimides, isoindolin-1-ones and isoindolines bearing aminobenzoic acids. The compounds showed fluorescence emission and high quantum yields in methanol solutions. The intramolecular charge in these donor-acceptor systems was found to significantly depend on electron-withdrawing substituents at the carboxylic acid position.
