1,2,4-Oxadiazole Topsentin Analogs with Antiproliferative Activity against Pancreatic Cancer Cells, Targeting GSK3β Kinase

Summary: A new series of topsentin analogs was efficiently synthesized by replacing the central imidazole ring of the natural lead with a 1,2,4-oxadiazole moiety. These compounds demonstrated significant antiproliferative activity against PDAC cells, reducing cell migration and promoting apoptosis. Further experiments suggest that these effects may be mediated through the modulation of epithelial-to-mesenchymal transition markers and GSK3 beta phosphorylation.
