Discovery of a novel series of indolylchalcone-benzenesulfonamide hybrids acting as selective carbonic anhydrase II inhibitors

Novel benzenesulfonamides aryl and arylsulfone conjugates adopting tail/dual tail approaches: Synthesis, carbonic anhydrase inhibitory activity and molecular modeling studies

Summary: New series of benzenesulfonamide and benzoic acid derivatives were designed and synthesized as carbonic anhydrase inhibitors using tail/dual tail approach. The benzenesulfonamide derivatives showed moderate to potent inhibitory activity with selectivity toward hCA II, while the benzoic acid analogues mostly exhibited no activity or weak activity. Fold-in-fold-out modifications were used to optimize the designed compounds.

Summary: This research reported the rational design, synthesis, carbonic anhydrases (CAs) inhibitory effects, and cytotoxicities of a series of 4-(3-(2-arylidenehydrazine-1-carbonyl)-5-(thiophen-2-yl)-1H-pyrazole-1-yl)benzenesulfonamides. The selective inhibitory effects of compound 18 on cancer-associated hCA IX and compound 17 on hCA XI were highlighted. Furthermore, some compounds showed comparable tumor-specificity to doxorubicin towards HSC-2 cancer cell line, with potential correlation to certain chemical descriptors.
