Risk Factors of Subsequent Central Nervous System Tumors after Childhood and Adolescent Cancers: Findings from the French Childhood Cancer Survivor Study

Neige Marie Yvanne Journy et al.

Summary: Childhood or adolescent cancer survivors undergoing cranial irradiation are at increased risk of developing subsequent primary neoplasms of the central nervous system (CNS), particularly meningiomas, and the risk is significantly associated with radiation dose. Monitoring patients with cranial irradiation should continue for at least 30 years post-treatment to ensure early detection and intervention. Identified risk factors, such as radiation dose and cumulative alkylating agents, can help inform long-term surveillance strategies for this population.

Insights from the clinical phenotype of subjects with Laron syndrome in Ecuador

Jaime Guevara-Aguirre et al.

Summary: The Ecuadorian cohort of subjects with LS has provided valuable insights into the genetic, clinical, and metabolic aspects of the condition. They exhibit unique characteristics such as a growth hormone receptor mutation, lower incidence of diabetes and cancer, and a metabolic phenotype of elevated but ineffective GH concentrations and efficient insulin levels. These findings suggest that the combination of low IGF-I signaling, decreased insulin resistance, and efficient serum insulin concentrations may contribute to the reduced incidence of diabetes and cancer in these individuals.

Mammary Tumors Growing in the Absence of Growth Hormone Are More Sensitive to Doxorubicin Than Wild-Type Tumors

Daniel D. Lantvit et al.

Summary: The study showed that mammary tumors induced in SDRs regressed significantly after doxorubicin treatment compared to WT tumors, indicating that reducing GH signaling may decrease the dose of doxorubicin needed to effectively treat breast cancer.

Sensitivity and specificity of the macimorelin test for diagnosis of AGHD

Jose M. Garcia et al.

Summary: The macimorelin test is effective for diagnosing AGHD and has an advantage over the ITT in avoiding hypoglycemia. The performance of macimorelin is not significantly influenced by age, BMI, or sex, indicating its robustness in AGHD diagnosis. The GH cutpoint of 5.1 ng/mL provides optimal specificity (96%) and high sensitivity (92%), showing good agreement with the ITT at the same cutpoint.

Safety of growth hormone (GH) treatment in GH deficient children and adults treated for cancer and non-malignant intracranial tumors-a review of research and clinical practice

Summary: The safety of GH replacement therapy in survivors of cancer and brain tumors remains debated, with conflicting results from studies, especially in different age groups. While GH replacement therapy is considered relatively safe in individuals with non-functioning brain tumors, the risk versus benefit profile in other patients remains unclear.
