Associations of Adiposity, Circulating Protein Biomarkers, and Risk of Major Vascular Diseases

Yuanjie Pang et al.

Summary: This subcohort study of 628 Chinese adults found that body mass index was positively associated with multiple protein biomarkers, some of which were also associated with the risk of cardiovascular disease. These findings suggest that proteins may partly explain the link between obesity and cardiovascular disease.

Biased agonism at chemokine receptors

Dylan Scott Eiger et al.

Summary: The human chemokine system consists of numerous chemokine ligands and receptors that regulate a wide range of cellular functions and biological processes. These interactions display biased agonism, where different ligands binding to the same receptor can lead to distinct biological effects through different pathways, modulated by changes in ligand, receptor, or specific cellular context. Enhanced understanding of biased agonism in the chemokine system may transform understanding of biased signaling across all GPCR subtypes and lead to development of pharmaceuticals with increased therapeutic efficacy and safer side effect profiles.

Carotid Plaque Inflammation Imaged by PET and Prediction of Recurrent Stroke at 5 Years

John Joseph McCabe et al.

Summary: The study aimed to determine the association between carotid plaque inflammation detected by F-18-fluorodeoxyglucose (18)FDG-PET and late (5-year) recurrent stroke. Results showed that baseline plaque SUVmax and SCAIL score independently predicted 5-year ipsilateral recurrent stroke. This suggests that plaque inflammation-related (18)FDG uptake could improve identification of recurrent ipsilateral stroke over a 5-year period.

Monocyte-Chemoattractant Protein-1 Levels in Human Atherosclerotic Lesions Associate With Plaque Vulnerability

Marios K. Georgakis et al.

Summary: The study revealed that MCP-1 levels in human atherosclerotic plaques are associated with vulnerability features, clinical manifestations, and increased risk of adverse vascular events. Higher levels of MCP-1 are linked to histopathologic and molecular markers of plaque vulnerability, clinical plaque instability, and risks of major adverse vascular events post plaque removal.

Efficacy and safety of low-dose colchicine in patients with coronary disease: a systematic review and meta-analysis of randomized trials

Aernoud T. L. Fiolet et al.

Summary: This meta-analysis indicates that low-dose colchicine reduces the risk of major adverse cardiovascular events, myocardial infarction, stroke, and the need for coronary revascularization in patients with coronary disease. There was no difference in all-cause mortality, with fewer cardiovascular deaths but more non-cardiovascular deaths.

Association of Circulating Monocyte Chemoattractant Protein-1 Levels With Cardiovascular Mortality A Meta-analysis of Population-Based Studies

Marios K. Georgakis et al.

Summary: This meta-analysis of 7 population-based cohort studies assesses the association between circulating monocyte-chemoattractant protein-1 levels and risk of cardiovascular disease and death. The findings suggest that higher baseline circulating MCP-1 levels are associated with higher risk of cardiovascular mortality over a long-term follow-up period exceeding 20 years, providing further support for the role of MCP-1 in cardiovascular disease development.

Biased action of the CXCR4-targeting drug plerixafor is essential for its superior hematopoietic stem cell mobilization

Astrid S. Jorgensen et al.

Summary: Plerixafor exhibits biased action by reversing the CXCL12 gradient across the bone marrow endothelium and showing superior therapeutic effect in HSC mobilization. In contrast, AMD11070, despite having superior antagonism, does not perform as effectively in HSC mobilization in vivo.

Summary: The study reveals a significant contribution of rare variants to common disease, with a large number of gene-phenotype associations detected through gene-based collapsing analysis that cannot be identified in single-variant association tests. Rare variants are also significantly enriched for loss-of-function-mediated traits and approved drug targets.
